Will it hurt an amp if you pull the power tubes and leave the amp turned on for awhile?

I do it for testing, a friend and i are just settling an argument

No.

Voltages will be slightly higher though. Make sure the filter caps are rated high enough.

its a fender bassman with 100uf 450v, and 20uf @ 500V

Yes.

To be 100% sure, you can pull the power tubes and measure the voltage across each cap. A couple of volts too many for a minute or two will not destroy them (say if you measure 505V on a 500V cap).
