It's virtually impossible to damage an amp by turning it on with a tube missing, certainly with a preamp/PI tube out. If an amp is cathode biased, it's a very bad idea to run it with less than the full power tube compliment, because they usually share a cathode resistor & pulling tubes rebiases the remaining tubes hotter, which can blow them. Fixed bias amps that run their power tubes at the limit of what the tube can take can be problematic, because less tubes means less current load on the PT and already high voltages can rise to dangerous levels.
