I work with an old ham operator who gave me his back up tube tester: Knight 400. Anyone have experience with this tester? It has a pull out manual with operating instructions. Is this piece of kit the equivalent of testing a semiconductor with a DVM?

I'm not familiar with that particular piece, but your comparison (DVM-semiconductor) is accurate in the following respect and something to keep in mind always.
A typical tube tester does not normally stress the tube like the actual circuit does.
So, if the tube tester says the tube is bad, it's bad.
If the tube tester says the tube is good, it could still be bad.
Don't misunderstand me. The tube tester is a great tool but, like any other tool, you have to be aware of it's limitations."I took a photo of my ohm meter... It didn't help." Enzo 8/20/22

yeah, a tube tester is OK at telling you a tube is bad, but not very good at telling you they are good. Which is what he just said.
They are OK at testing emission, so you can compare two tubes to some extent.Education is what you're left with after you have forgotten what you have learned.
